摘要

中文摘要 配伍作為復方中藥物的核心關系,是復方發揮療效的關鍵,其科學性是闡明方劑作用機制的基礎。中藥復方配伍機制研究長期以來主要圍繞"化學成分-藥效學-藥動學"3個角度分別展開。補腎經方青娥方由杜仲(鹽炒)、補骨脂(鹽炒)、核桃仁(炒)、大蒜(蒸熟)四味藥組成,具有雌激素樣作用,且以全方效果最佳,但其配伍增效機制尚不明確。本研究擬以青娥方為研究對象,從補骨脂入手,圍繞青娥方的雌激素樣作用開展補骨脂與各組方藥味配伍前后的藥效學比較研究;同時研究青娥方全方及各配伍藥物組合物中補骨脂相關活性成分的吸收、分布、代謝的變化;采用代謝組學、藥代動力學方法發現表征配伍前后藥效學變化的生物標示物和化學標示物,從藥效學和藥動學相互作用角度闡明青娥方的配伍機制。研究結果可為青娥方配伍機制的闡明提供理論依據,也可為其它中藥復方配伍機制研究提供借鑒。
英文摘要 Compatibility theory is the key problem for the interpretation the function mechanism of traditional Chinese compound preparations. Most herb-herb interactions are mediated by pharmacodynamic and/or pharmacokinetic mechanisms. Qing'e Fomula is a famous compound preparation which consists of four ingredients, Eucommiae Cortex (Duzhong, stir-baked with salt), Psoraleae Fructus (stir-baked with salt), Juglandis Semen (stir-baked) and Bulbus Allii(steamed). Previously studies show that Qing'e Fomula has estrogenic activities, but its compatibility synergia mechanism is still unclear. In this study,compatibility mechanism studies of Qing'e Fomula based on herb-herb interaction between Posoraleae Fructus and other ingredients will be carried on. In Vivo and in vitro studies on the pharmacodynamic and pharmacokinetic interactions between the four herbs will be investigated. Pharmacokinetics and metabonomics method will be used to find and identify the related biological markers, pathological indicators and determinate the biochemical.The research results is prospected to provide theoretical basis for explaination of the compatibility mechanism of Qing'E formula on the estrogenic activity, and to have important guiding significance for the research of the compatibility mechanism of other traditional Chinese compound preparations.
